目的探讨化浊颗粒对非酒精性脂肪肝病(NAFLD)大鼠模型血脂代谢的干预作用。方法将50只大鼠随机分为空白组、模型组、罗格列酮组、化浊颗粒高、低剂量组各10只,空白组以标准饲料喂养;其余大鼠以高脂饲料喂养8周建立NAFLD大鼠模型,罗格列酮组给予罗格列酮片0.36mg/kg灌胃,化浊颗粒高、低剂量组分别给予化浊颗粒2.025、0.675g/kg灌胃,模型组予等量生理盐水灌胃,均每日1次。治疗4周后观察各组大鼠体重、肝指数、总胆固醇(TC)、甘油三酯(TG)、肿瘤坏死因子(TNF-α)、肝细胞脂肪变性的变化。结果化浊颗粒高剂量组大鼠治疗后体重较治疗前降低(P<0.01);与模型组相比,化浊颗粒高剂量组及罗格列酮组肝指数、TG、TC、TNF-α均降低(P<0.05或P<0.01)。结论化浊颗粒可降低NAFLD大鼠TG、TC、TNF-α,从而改善NAFLD大鼠的血脂代谢异常和病理改变。
Objective To investigate the intervention of Huazhuo granule on lipid metabolism in non-alcoholic fatty liver disease (NAFLD) rat model. Methods Fifty rats were randomly divided into blank group, model group, rosiglitazone group, high and low dose of Huazhuo Granules group, 10 rats in each group. The blank group was fed with standard diet. The rest rats were fed with high fat diet for 8 weeks NAFLD rat model was established. Rosiglitazone group was administrated rosiglitazone 0.36mg / kg intragastrically. The mice in high and low dose groups were given intragastric administration of Huazhuo granules 2.025 and 0.675g / kg respectively. The model group was given The amount of saline irrigation, all day 1. After 4 weeks of treatment, the changes of body weight, liver index, total cholesterol (TC), triglyceride (TG), tumor necrosis factor (TNF-α) and fatty degeneration of hepatocytes were observed. Results Compared with the untreated group, the weight of high-dose Huazhuo Granules group was significantly lower than that before treatment (P <0.01). Compared with the model group, the indexes of liver index, TG, TC, TNF-α (P <0.05 or P <0.01). Conclusion Huazhuo Granules can reduce TG, TC and TNF-α in NAFLD rats, and thus improve blood lipid metabolism and pathological changes in NAFLD rats.
