目的:研究牻牛儿苗亲缘关系和遗传多样性。方法:23个样品进行简单序列重复区间(ISSR)分析,利用NTSYS软件计算材料间遗传相似系数,并用UPGMA方法聚类,构建亲缘关系系统图。结果:从100条ISSR引物中筛选出14条多态性明显、反应稳定的引物,在23份供试材料DNA中共扩增出185条谱带,其中多态性条带142条,占76.8%。材料间ISSR标记遗传相似性系数(GS)在0.458~0.916。聚类分析结果显示,所有供试材料均可区分开,并聚为6组。结论:分子标记研究结果与牻牛儿苗的地理位置、形态性状存在显著的相关性,较好地揭示了牻牛儿苗种源间的遗传关系,可为牻牛儿苗资源划分和良种选育提供科学依据。
Objective: To study the genetic relationship and genetic diversity of geranium seedlings. Methods: 23 samples were analyzed by simple sequence repeat interval (ISSR), the genetic similarity coefficient was calculated by NTSYS software, and the phylogenetic relationship was constructed by UPGMA clustering. Results: Of the 100 ISSR primers, 14 were identified with stable polymorphisms and 185 stable bands. A total of 185 bands were amplified from 23 DNA samples, of which 142 were polymorphic, accounting for 76.8% . The genetic similarity coefficient (GS) of ISSR markers was between 0.458 and 0.916. Cluster analysis showed that all the tested materials could be distinguished and clustered into 6 groups. Conclusion: The results of molecular markers showed significant correlation with the geographical location and morphological traits of gentian seedlings, revealing the genetic relationship between germplasm resources of geranium seedlings better, Education provides a scientific basis.
