目的:研究新鲜何首乌幼茎、幼叶及块根的超微结构。方法:采用ODO扫描电镜法和冷冻蚀刻法行电镜观察。结果:发现叶表皮细胞游离面具有角质纹理,气孔由周细胞和中心细胞组成;幼茎韧皮部细胞核孔散在,膜蛋白颗粒散在或密集成片;块根细胞质内淀粉粒的表面附有大小不一的微粒。结论:将ODO法及冷冻蚀刻法引入中草药学结构研究,可获得比光学显微镜及超薄切片透射电镜法更好的优势,并能发现更多植物细胞的三维微细结构。
Objective: To study the ultrastructure of young stems, young leaves and roots of fresh root of Polygonum multiflorum. Methods: ODO scanning electron microscopy and frozen etching were used to observe the electron microscopy. RESULTS: It was found that the free surface of leaf epidermal cells had a keratinous texture, the stomata consisted of pericytes and central cells; the nucleus pores of the young phloem were scattered, and the membrane protein particles were scattered or densely integrated into the sheet; the surface of the starch granules in the cytoplasm of the tuberous root had different sizes. particle. Conclusion: The introduction of ODO method and frozen etching method into the structure of Chinese herbal medicine can obtain better advantages than optical microscopy and ultrathin section transmission electron microscopy, and can discover more three-dimensional microstructure of plant cells.
