以感病的朝天委陵菜(Potentilla supine)叶片为试材,采用病原学及分子生物学方法,研究了河南商丘地区朝天委陵菜白粉病病原菌的系统进化关系。结果表明:该病原菌分生孢子梗直立,圆柱形,简单无分枝,大小为(90~180)μm×(10~17)μm,分生孢子透明,椭圆形,有明显的纤维体,大小为(17~33)μm×(13~18)μm,4~6个串生;病原菌接种叶片病症与自然状态一致,白色菌丝体部分覆盖上层叶表面,随着病菌的发展,菌丝蔓延至覆盖整个叶子表面;对其核糖体DNA内转录间隔区(ITS)序列进行PCR扩增、测序获得563bp序列(GenBank登录号KR049083)。经MEGA 3.1软件分析,其与来自佩兰(Herb eupatorii)棕丝单囊白粉菌(Podosphaere fusca)的序列(GenBank登录号JX548297)聚为一支,而与来自同属另4个种的ITS序列亲缘关系较远。研究表明,河南省商丘地区的朝天委陵菜白粉菌为棕丝单囊白粉菌(Podosphaera fusca)。
The susceptible leaves of Potentilla supine were used to study the phylogenetic relationship of Potentilla supine leaves from Potentilla aestivum in Shangqiu, Henan Province, using etiological and molecular methods. The results showed that the conidiophores of the pathogen were upright, cylindrical and simple without branches. The size of the conidiophores was (90-180) μm × (10-17) μm. The conidia were transparent and oval with obvious fibrous body, size (17 ~ 33) μm × (13 ~ 18) μm, and 4 ~ 6 strings. The pathogen inoculated leaves were consistent with their natural state. The white mycelium partially covered the surface of upper leaf. With the development of germs, mycelium spread To cover the entire leaf surface. The ITS sequence of ribosomal DNA was amplified by PCR and sequenced to obtain a 563 bp sequence (GenBank Accession No. KR049083). It was integrated with the sequence of Podosphaere fusca from Herb eupatorii (GenBank accession number JX548297) by MEGA 3.1 software and was related to ITS sequences from the other 4 species Far. Studies have shown that the Aspergillus tinctorius Potentilla in Henan Shangqiu region is Podosphaera fusca.
