目的观察油酸致大鼠急性呼吸窘迫综合症(ARDS)模型中肺泡II型细胞cAMP、cGMP水平的变化及特布他林的作用。方法随机将SD大鼠分为对照组、ARDS组、特布他林组。用放射免疫分析法测定肺泡II型上皮细胞内cAMP、cGMP水平;应用重力法测定血管外肺水含量(EVLW)。结果ARDS组大鼠肺泡II型上皮细胞内cAMP明显降低,cGMP水平升高,EVLW明显增多。给予特布他林治疗后,EVLW下降,肺泡II型细胞内cAMP含量升高,cGMP变化不明显。结论油酸致ARDS时,肺水清除能力下降。特布他林可通过提高肺泡II型细胞内cAMP水平提高肺氷清除能力。肺泡II型细胞内cAMP、cGMP水平变化参与急性肺损伤的分子生物学发病机制。
Objective To observe the changes of cAMP and cGMP levels in type 2 alveolar cells in acute respiratory distress syndrome (ARDS) induced by oleic acid and the effect of terbutaline. Methods SD rats were randomly divided into control group, ARDS group and terbutaline group. Radioimmunoassay was used to determine the level of cAMP and cGMP in alveolar type II epithelial cells. The extravascular lung water content (EVLW) was measured by gravimetry. Results In ARDS group, the level of cAMP in alveolar type II cells was significantly decreased, the level of cGMP was increased, and the level of EVLW was significantly increased. After treatment with terbutaline, the EVLW decreased, the content of cAMP in alveolar type II cells increased, and the change of cGMP was not obvious. Conclusion When oleic acid induced ARDS, lung water scavenging ability decreased. Terbutaline improves lung ice clearance by increasing cAMP levels in alveolar type II cells. Alteration of cAMP and cGMP levels in alveolar type II cells is involved in the molecular pathogenesis of acute lung injury.
