目的:分析中国人血清中庚型肝炎病毒(HGV或GBV-C)NS3区部分基因序列。方法:从个体供血者及肝硬化患者血清中,通过逆转录合成cDNA,设计特异性引物进行多聚酶链式聚合反应,将产物克隆于载体上,采用双脱氧核苷酸链终止法进行双向测序。结果:分离出的5株NGV毒株的NS3区部分核苷酸序列之间同源性从85%到98%,与国外已发表的HGV序列比较,同源性在79%~91%。推测的其编码氨基酸序列,5株之间100%一致,与国外已报告的HGV比较,同源性为94.9%~100%。结论:从中国人血清中分离出5株HGV序列,其NS3区部分核苷酸存在着一定的变异性,但其编码的氨基酸非常保守。
OBJECTIVE: To analyze the partial gene sequences of NS3 region of hepatitis G virus (HGV or GBV-C) in Chinese human serum. Methods: The cDNAs were synthesized by reverse transcription from the blood serum of individual blood donors and patients with cirrhosis, and specific primers were designed for polymerase chain reaction. The products were cloned into vector and sequenced by dideoxynucleotide chain termination method. Results: The homology of partial NS3 nucleotide sequences of the 5 strains of NGV isolated from 85% to 98% were 79% -91% homologous with published HGV sequences. The deduced amino acid sequence of the deduced amino acid sequence was 100% homologous among the 5 strains. The homology was 94.9% -100% with the reported foreign HGV. CONCLUSION: Five HGV sequences isolated from Chinese serum have some mutations in some of the NS3 nucleotide sequences, but the amino acids are very conserved.
