目的:探讨SD大鼠柔红霉素(DNR)诱导性亚急性心肌损伤大鼠模型的建立和评价方法。方法:SD大鼠随机分成2组:对照组(n=15)和DNR组(n=15)。DNR组腹腔注射柔红霉素3.5 mg·kg-1,每周一次,连续4周。对照组腹腔注射等体积生理盐水。检测心电、心脏结构和功能、心肌病理学改变及血清心肌肌钙蛋白T(cTnT)及肌酸激酶同工酶(CK-MB)水平的变化。结果:与对照组比较,心电图QRS波电压下降,ST段抬高。EF值下降。HE染色结果显示心肌结构紊乱,细胞间隙增宽。血清CK-MB和cTnT水平升高(P<0.05)。结论:腹腔注射DNR可成功诱导亚急性心肌损伤大鼠模型,可通过检测心电、血浆cTnT和CK-MB、心功能和结构变化评价。
Objective: To investigate the establishment and evaluation of rat model of daunorubicin (DNR) -induced subacute myocardial injury in SD rats. Methods: SD rats were randomly divided into 2 groups: control group (n = 15) and DNR group (n = 15). DNR group intraperitoneally injected daunorubicin 3.5 mg · kg-1 once a week for 4 weeks. The control group was injected intraperitoneally with equal volume of normal saline. The changes of ECG, cardiac structure and function, changes of cardiomyopathology and levels of cardiac troponin T (cTnT) and creatine kinase MB (CK-MB) were detected. Results: Compared with the control group, ECG QRS wave voltage decreased, ST segment elevation. EF value decreased. HE staining showed that myocardial structure disorder, widening of the cell gap. Serum CK-MB and cTnT levels increased (P <0.05). CONCLUSION: Intraperitoneal injection of DNR can successfully induce subacute myocardial injury in rats, which can be evaluated by detecting ECG, plasma cTnT and CK-MB, cardiac function and structural changes.
