选用5个品种的彩色辣椒,研究不同品种、不同培养方式、不同子叶生长发育时期对愈伤组织诱导和形成的影响,以及不定芽分化情况。结果表明:在MS固体培养基中添加6-BA5.0mg/l时愈伤组织诱导频率明显高于其它激素组合;进一步试验表明以半固体培养方式(琼脂含量为0.25%)和无菌苗子叶处于初展期时进行培养愈伤组织诱导率明显提高,且质量好;将所获得的愈伤组织进一步培养不定芽分化率高达100%。
Five varieties of colored pepper were selected to study the effects of different varieties, different culture methods and different cotyledon growth and development stages on the induction and formation of callus and the differentiation of adventitious buds. The results showed that the induction frequency of callus was significantly higher than that of other hormones when 6-BA5.0mg / l was added into MS solid medium. Further experiments showed that the induction rate of callus was higher in semi-solid culture (agar content 0.25%) and sterile seedling cotyledons The callus induction rate was significantly improved and the quality was good when the callus was cultured at the initial stage. The callus obtained was further cultured and the differentiation rate of adventitious buds was as high as 100%.
