目的从下丘脑-垂体-甲状腺(HPT)轴角度研究金匮肾气汤对肾阳虚小鼠、六味地黄汤对肾阴虚小鼠的影响,探求金匮肾气汤与六味地黄汤不同的补肾作用机制。方法取昆明小鼠50只,随机分为5组(每组10只):空白组、肾阳虚模型组、肾阴虚模型组、金匮肾气汤组、六味地黄汤组;每天灌胃氢化可的松注射液,制备小鼠肾阳虚、肾阴虚模型;测定体重、环磷酸腺苷(cAMP)、环磷酸鸟苷(cGMP)、甲状腺激素(TSH)、三碘甲腺原氨酸(T3)、甲状腺素(T4)含量,观察甲状腺组织形态学改变。结果肾阳虚模型小鼠体重、血浆cAMP含量、cAMP/cGMP比值及血清TSH、T3、T4含量降低;肾阴虚模型小鼠体重下降,血浆cAMP含量、cAMP/cGMP比值及血清TSH、T3、T4含量升高;金匮肾气汤能上调肾阳虚模型小鼠体重、血浆cAMP含量、cAMP/cGMP比值及血清TSH、T3、T4含量;六味地黄汤能上调肾阴虚模型小鼠体重,下调血浆cAMP含量、cAMP/cGMP比值及血清TSH、T3、T4含量;两方均能修复受损甲状腺组织。结论金匮肾气汤改善肾阳虚证的作用机制之一可能与其升高HPT轴中垂体分泌的TSH,甲状腺分泌的T3、T4有关,六味地黄汤改善肾阴虚证的作用机制则反之。
Objective To study the effect of Jinkui Shenqi Decoction on kidney-yin deficiency mice and Liuweidihuang decoction on kidney-yin deficiency mice from the angle of hypothalamus-pituitary-thyroid (HPT) axis, and to find out that Jingui Shenqi Decoction is different from Liuwei Dihuang Decoction Bushen mechanism. Methods Fifty Kunming mice were randomly divided into 5 groups (10 in each group): blank group, Shen Yang deficiency model group, Shen Yin deficiency group, Jingui Shenqi Decoction group and Liuweidihuang decoction group; Hydrocortisone injection was used to prepare the model of kidney-yang deficiency and kidney-yin deficiency in mice. The body weight, cAMP, cGMP, thyroid hormones (TSH), triiodothyronine Acid (T3), thyroxine (T4) content, observe the histological changes of thyroid tissue. Results The body weight, plasma cAMP, cAMP / cGMP and serum TSH, T3 and T4 in kidney-yang deficiency model mice were decreased. The body weight, plasma cAMP, cAMP / cGMP and serum TSH, T4 decoction could increase the content of cAMP, cAMP / cGMP, TSH, T3 and T4 in mice with deficiency of kidney yang. Liuweidihuang Tang can up-regulate the body weight, Decreased plasma cAMP content, cAMP / cGMP ratio and serum TSH, T3, T4 content; both can repair damaged thyroid tissue. Conclusion Jingui Shenqi Decoction can improve the mechanism of deficiency of Kidney-Yang deficiency possibly by increasing TSH secreted by pituitary gland in HPT axis and T3 and T4 secretion by thyroid gland. The mechanism of Liuweidihuang Decoction in improving kidney-yin deficiency is opposite.
