目的:观察丙型肝炎病毒(HCV)核心蛋白DNA疫苗诱导BALB/c小鼠体液免疫应答的效力,为研制应用于人类的HCVDNA疫苗提供实验依据。方法:将全长HCV核心蛋白基因插入真核表达质粒载体pcDNA3.1,构建HCV核心蛋白重组表达质粒pcDNA3.1HCVcore,肌注BALB/c小鼠,以ELISA法检测小鼠血清HCV抗体。以此重组质粒转染小鼠NIH3T3细胞,以HCV抗体阳性小鼠血清为捕获抗体进行Westernblot,检测DNA疫苗编码的HCV核心蛋白的表达。结果:30%(3/10)免疫小鼠产生HCV核心蛋白抗体,该抗体能检测到重组质粒在NIH3T3细胞表达的HCV核心蛋白。结论:HCV核心蛋白DNA疫苗能诱导BALB/c小鼠体液免疫应答
Objective: To observe the efficacy of Hepatitis C virus (HCV) core protein DNA vaccine in inducing humoral immune response in BALB / c mice and provide experimental evidence for the development of human HCVDNA vaccine. Methods: The full-length HCV core protein gene was inserted into the eukaryotic expression plasmid vector pcDNA3.1 to construct HCV core protein recombinant plasmid pcDNA3.1 HCVVore, BALB / c mice were intramuscularly injected, and the serum anti-HCV antibodies were detected by ELISA. The recombinant plasmids were transfected into mouse NIH3T3 cells and HCV antibody-positive mouse sera were used as the capture antibody for Western blotting to detect the expression of HCV core protein encoded by the DNA vaccine. RESULTS: Thirty (3/10) immunized mice developed HCV core protein antibodies that detected the HCV core protein expressed by recombinant plasmids in NIH3T3 cells. Conclusion: HCV core DNA vaccine can induce humoral immune response in BALB / c mice
