为了解白介素 1β(IL 1β)、肿瘤坏死因子α(TNF α)对大鼠肝星状细胞间质胶原酶 (MMP13 )基因表达的影响 ,在培养的肝星状细胞系中加入IL 1β、TNF α,于不同的时间点收集细胞 ,提取总RNA ,用逆转录定量PCR方法测定MMP13 的基因表达水平。结果表明 ,IL 1β组肝星状细胞MMP13 基因表达水平在 8、2 4、4 8h三个时间点均明显高于对照组 ,2 4h达高峰 ,为对照组的 3倍 ;TNF α组肝星状细胞MMP13 的基因表达水平在 8、2 4、4 8、72h均明显高于对照组 ,4 8h达高峰 ,为对照组的 4倍。上述结果提示IL 1β、TNF α可增强肝星状细胞MMP13 基因的表达
To investigate the effects of interleukin 1β (IL 1β) and tumor necrosis factor α (TNFα) on the gene expression of rat interstitial collagenase (MMP13) in rat hepatic stellate cells, IL 1β, TNF α, cells were collected at different time points, total RNA was extracted, and the gene expression level of MMP13 was determined by reverse transcription quantitative PCR. The results showed that the expression of MMP13 gene in hepatic stellate cells of IL 1β group was significantly higher than that of the control group at 8,2,4,48 h and reached the peak at 24 h, which was three times of that of the control group. The gene expression levels of MMP13 in the somatic cells were significantly higher than those in the control group at 8, 2, 4, 8 and 72 hours, reaching the peak at 48 hours, which was 4 times of the control group. The above results suggest that IL 1β and TNFα can enhance the expression of MMP13 in hepatic stellate cells
