目的探讨围产期低剂量三丁基锡(TBT)暴露对雌性昆明小鼠生长发育及血清雌激素的影响。方法将清洁级健康成年妊娠昆明小鼠随机分为100、10、1μg/kgTBT染毒组和对照组(玉米油),每组6只。从妊娠第6天起,采用经口灌胃方式进行染毒,每天1次,染毒剂量为1ml/kg,直至哺乳期结束。出生后第49天,每组随机抽取10只雌性仔鼠,称重后取血,分离子宫及卵巢,称重,计算子宫及卵巢的脏器系数。采用放射免疫法测定血清中雌二醇的浓度。结果与对照组比较,各TBT染毒组雌性仔鼠睁眼时间延长,阴道张开时间提前,差异均有统计学意义(P<0.05或P<0.01);且随着TBT染毒剂量的增加,雌性仔鼠睁眼时间呈延长趋势。各TBT染毒剂量组雌性仔鼠体重、子宫重量及其脏器系数间比较,差异均无统计学意义(P>0.05)。与对照组比较,10μg/kgTBT染毒组雌性仔鼠血清中雌二醇的浓度和卵巢重量及其脏器系数升高,差异均有统计学意义(P<0.01);且仔鼠血清雌二醇水平随着染毒剂量的增加呈递增趋势。结论围产期低剂量TBT暴露对雌性仔鼠有内分泌干扰作用,影响其生长发育,导致血清雌激素水平升高。
Objective To investigate the effect of low dose TBT exposure on the growth and serum estrogen in female Kunming mice. Methods Healthy pregnant Kunming mice of healthy grade were randomly divided into 100,10,1μg / kg TBT group and control group (corn oil), 6 mice in each group. From the 6th day of gestation, the rats were orally administered by oral gavage once a day at a dose of 1ml / kg until the end of lactation. On the 49th day after birth, 10 female offspring were randomly selected from each group. Blood was weighed, uterus and ovaries were separated, weighed and the organ coefficients of uterus and ovary were calculated. Serum estradiol concentration was determined by radioimmunoassay. Results Compared with the control group, the female pups in TBT exposure group had longer eyes open time and earlier vaginal opening time (P <0.05 or P <0.01), and with the increase of TBT dose , Female pups eyes open time was extended. There was no significant difference in body weight, uterine weight and organ coefficient among female TBI dose groups (P> 0.05). Compared with the control group, the concentration of estradiol and the weight of ovary and the organ coefficient of the female offspring in 10 μg / kg TBT treatment group were significantly increased (P <0.01), and the serum estradiol Alcohol levels increased with the increase of exposure dose. Conclusions Perinatal low-dose TBT exposure has endocrine disrupting effects on female offspring, which affects its growth and development and leads to elevated serum estrogen levels.
