从多粘菌素B处理的豚鼠腹腔渗出液中,分离嗜酸性粒细胞(Eos)用超声粉碎仪破坏细胞,离心后收集含Eos成分的上清液,给豚鼠经气管插管雾化喷入。60min后豚鼠对组胺的气道反应性PC20值显著降低(P<0.05)。Eos成分吸入处理组和正常对照组支气管肺泡灌洗液(BALF)中的细胞总数、Eos绝对计数差异均无显著意义(P>0.05)。但BALF细胞中上皮细胞的百分比显著增多(P<0.001),并且与60min时PC20值的降低呈显著正相关(r=0.84,P<0.05)。以上结果提示Eos可能通过其细胞成分造成气道上皮损伤诱导气道高反应性。
From polymyxin B-treated guinea pig peritoneal exudate, eosinophils (Eos) were isolated and the cells were disrupted with an ultrasonic disintegrator. After centrifugation, supernatants containing Eos components were collected and administered to guinea pigs for intubation Into. After 60 minutes, the airway responsive PC20 value of histamine was significantly decreased in guinea pigs (P <0.05). There was no significant difference in the total count of Eos and count of Eos in bronchoalveolar lavage fluid (BALF) between the Eos composition inhalation group and the normal control group (P> 0.05). However, the percentage of epithelial cells in BALF cells was significantly increased (P <0.001), and was positively correlated with the decrease of PC20 value at 60 min (r = 0.84, P <0.05). The above results suggest that Eos may induce airway hyperresponsiveness through airway epithelial damage caused by its cellular components.
