目的:研究姜黄素和顺铂联用抑制雄激素非依赖性前列腺癌细胞株PC-3增殖的影响。方法:MTT比色法检测细胞生长活性,流式细胞仪测定细胞周期时相变化及检测细胞凋亡,透射电镜观察细胞超微结构变化。结果:姜黄素能显著抑制雄激素非依赖性前列腺癌细胞的体外生长,呈时间与剂量依赖性;细胞周期主要阻滞于G2/M期,部分细胞出现凋亡形态学改变。顺铂和不同浓度姜黄素联合可显著抑制PC-3细胞增殖。姜黄素和顺铂在一定程度上均可诱导PC3细胞凋亡,两种药物联用后,诱导细胞凋亡作用增强。结论:姜黄素可与顺铂协同抑制人前列腺癌细胞株PC3的增殖。
OBJECTIVE: To investigate the effect of curcumin and cisplatin on the proliferation of androgen-independent prostate cancer cell line PC-3. Methods: MTT colorimetric assay was used to detect the cell growth activity. Flow cytometry was used to determine the cell cycle phase change and cell apoptosis was detected. Transmission electron microscopy was used to observe the changes of cell ultrastructure. RESULTS: Curcumin could significantly inhibit the growth of androgen-independent prostate cancer cells in vitro and in a dose-dependent manner. The cell cycle arrested mainly in the G2/M phase, and some cells showed apoptotic morphological changes. The combination of cisplatin and different concentrations of curcumin could significantly inhibit the proliferation of PC-3 cells. Curcumin and cisplatin can induce apoptosis of PC3 cells to a certain extent. After the combination of the two drugs, the induction of apoptosis is enhanced. Conclusion: Curcumin can synergize with cisplatin to inhibit the proliferation of human prostate cancer cell line PC3.
