葛根药材HPLC指纹图谱研究

目的研究葛根药材的指纹图谱,从整体上控制葛根药材的质量。方法采用HPLC法,用二级管阵列进行检测,以葛根黄酮类成分分离效果为评价指标,筛选流动相,检测波长及提取方法,并进行方法学考察。结果以30%乙醇回流提取1小时,用乙腈-0.1%乙酸梯度洗脱效果较佳,检测波长为250nm。葛根药材在组分上相似度较好,各组分的含量差别不大。结论经统计分析,该法稳定,重现性好,简便可行,适用于葛根药材的质量控制。

Objective To study the fingerprint of Radix Puerariae and to control the quality of Radix Puerariae as a whole. Methods HPLC method was used to detect with a diode array. The separation effect of flavonoids from Pueraria lobata was evaluated, and the mobile phase, detection wavelength and extraction method were screened. The results were extracted with 30% ethanol reflux for 1 hour, and the gradient elution with acetonitrile-0.1% acetic acid was better. The detection wavelength was 250 nm. Radix Puerariae has similarity in the components and the differences in the contents of the components are not significant. Conclusion According to statistical analysis, the method is stable, reproducible, simple and feasible, and suitable for the quality control of Radix Puerariae.
