目的:用HPLC-ELSD法直接测定复方甘草酸铵注射液中2种未衍生氨基酸的含量。方法:使用Agilent-ODS C18(4.6 mm×250 mm,5μm)色谱柱,流动相A(乙腈)-B(5.0 mmol.L-1七氟丁酸的0.7%三氟醋酸溶液),梯度洗脱(0~5 min,0%A;5~10 min,15%A;10~25 min,35%A),ELSD漂移管温度80℃,氮气流量2.0 L.min-1,流速0.8 mL.min-1,柱温35℃,进样10μL。结果:甘氨酸进样量在4.858~29.145μg,盐酸半胱氨酸进样量在0.386~2.316μg时,其对数值与峰面积对数值线性关系良好,回收率分别为97.43%,96.99%。结论:测定方法简便,结果准确,可用于复方甘草酸铵注射液中2种氨基酸的测定。
OBJECTIVE: To determine the content of two non-derivatized amino acids in compound glycyrrhizinate injection by HPLC-ELSD. Methods: A mobile phase A (acetonitrile) -B (0.7 mmol trifluoroacetic acid solution of 5.0 mmol.L-1 heptafluorobutyric acid) was used on a Agilent-ODS C18 column (4.6 mm × 250 mm, 5 μm) (0 to 5 min, 0% A; 5 to 10 min, 15% A; 10 to 25 min, 35% A), the ELSD drift tube temperature was 80 ℃, the nitrogen flow rate was 2.0 L.min-1 and the flow rate was 0.8 mL.min -1, column temperature 35 ℃, injection of 10μL. Results: The linearity of logarithmic value and peak area of glycine was 4.858-29.145μg and the cysteine hydrochloride sample was 0.386-2.316μg. The recoveries were 97.43% and 96.99%, respectively. Conclusion: The method is simple and accurate and can be used for the determination of two kinds of amino acids in compound glycyrrhizinate injection.
