目的:采用免疫组化法检测人牙龈成纤维细胞(HGFs)肿瘤坏死因子受体相关因子-6(TRAF-6)的表达及LPS的影响。方法:采用原代培养的人牙龈成纤维细胞,取3~8代细胞以LPS梯度浓度干预,24 h后观察干预后人牙龈成纤维细胞形态学的变化,并采用免疫组织化学的方法检测TRAF6在各组细胞中的表达。结果:TRAF-6在正常的HGFs中呈阴性表达,随LPS干预浓度由0.1μg/ml增加到100μg/ml,TRAF-6表达有先逐渐增高然后又降低的趋势。结论:TRAF-6在LPS引发的HGFs炎症损伤中发挥重要的作用。
OBJECTIVE: To detect the expression of tumor necrosis factor receptor associated factor-6 (TRAF-6) and the effect of LPS on human gingival fibroblasts (HGFs) by immunohistochemistry. Methods: Human gingival fibroblasts cultured in primary culture were used. After 3-8 passages of cells were treated with LPS gradient concentration, the morphological changes of human gingival fibroblasts were observed 24 hours later. The expression of TRAF6 was detected by immunohistochemistry Expression in each group of cells. Results: TRAF-6 was negatively expressed in normal HGFs. The TRAF-6 expression increased gradually and then decreased with the increase of LPS concentration from 0.1μg / ml to 100μg / ml. Conclusion: TRAF-6 plays an important role in LPS-induced inflammatory injury of HGFs.
