目的:探讨eIF4E在喉鳞癌(LSCC)中的表达及其与LSCC的关系。方法:采用免疫组织化学法检测52例蜡块LSCC组织、20例喉黏膜不典型增生组织、30例喉良性增生组织及10例正常喉黏膜组织中eIF4E基因蛋白的表达情况;用Western-blot法检测20例新鲜LSCC标本的癌中心区(TC)、过渡区(TR)及病理证实的无癌区(TF)中eIF4E的表达情况。结果:免疫组织化学法检测示:eIF4E在LSCC中表达的阳性率为100.0%,显著高于对照组。Western-blot法检测显示:eIF4E在TF、TR及TC的表达水平呈进行性增高。结论:过度表达的eIF4E是LSCC恶性改变及判断其恶性程度的一个分子标志,可为LSCC的基因治疗提供一定的理论基础。
Objective: To investigate the expression of eIF4E in laryngeal squamous cell carcinoma (LSCC) and its relationship with LSCC. Methods: Immunohistochemical method was used to detect the expression of eIF4E protein in 52 cases of LSCC, 20 cases of laryngeal mucosa atypical hyperplasia, 30 cases of laryngeal benign hyperplasia and 10 cases of normal laryngeal mucosa. Western-blot The expression of eIF4E was detected in 20 cases of fresh LSCC specimens from the center of tumor (TC), transitional zone (TR) and pathologically confirmed non-cancerous zone (TF). Results: Immunohistochemistry showed that the positive rate of eIF4E expression in LSCC was 100.0%, which was significantly higher than that of the control group. The results of Western-blot showed that the expression of eIF4E in TF, TR and TC increased progressively. Conclusion: Overexpression of eIF4E is a molecular marker of malignant transformation of LSCC and its malignancy, which may provide some theoretical basis for gene therapy of LSCC.
