目的探讨中草药对麻黄素成瘾动物的影响。方法采用递增剂量腹腔连续注射麻黄素溶液(2.0g/L、3.0g/L和4.0g/L)15d,建立麻黄素依赖动物模型,再分别给予递增剂量(20.0g/L、30.0g/L、40.0g/L)的黄芪、川芎及中药复方液进行灌胃,分别于灌胃中药5、10、15d用分光光度法检测肾组织超氧化物岐化酶(SOD)、过氧化氢酶(CAT)活性及丙二醛(MDA))含量和血清尿素氮含量,用免疫组织化学法检测肾组织中Bax蛋白和转化生长因子-β1(TGF-β1)的表达,用生物显微技术观察肾组织结构的变化。结果实验对照组小鼠肾组织SOD和CAT活性降低,MDA含量升高,肾组织Bax蛋白和TGF-β1的阳性表达增强,血清中尿素氮含量升高,肾组织严重受损;黄芪组、川芎组及中药复方组小鼠肾组织MDA含量又降低(P<0.05或P<0.01),SOD和CAT活性又升高(P<0.05或P<0.01),血清中尿素氮含量降低,肾组织损伤变性明显减轻,肾组织Bax蛋白和TGF-β1的阳性表达强度减弱。结论黄芪、川芎及中药复方可提高肾组织细胞的抗氧化能力,促进组织细胞损伤修复。
Objective To investigate the effects of Chinese herbal medicine on ephedrine-addicted animals. Methods Ephedrine-dependent animal model was established by continuous injection of escalating dose of ephedrine solution (2.0g / L, 3.0g / L and 4.0g / L) for 15d, and then treated with escalating doses of 20.0g / L and 30.0g / L , 40.0g / L) astragalus root, Chuanxiong rhizome and traditional Chinese medicine compound liquid respectively. Astragalus membranaceus, (P <0.05 or P <0.01), the activity of SOD and CAT increased again (P <0.05 or P <0.01), the level of serum urea nitrogen decreased, and the level of renal tissue injury Degeneration was significantly reduced, the expression of Bax protein and TGF-β1 in renal tissue decreased. Conclusion Astragalus, Chuanxiong and traditional Chinese medicine compound can improve renal cell antioxidant capacity and promote tissue injury repair.
