目的研究大量元素和植物生长物质对怀牛膝愈伤组织生长及怀牛膝多糖量的影响。方法以怀牛膝无菌苗的叶片、茎段为材料,采用正交试验设计方法研究培养基中大量元素和植物生长物质6-BA、IBA、NAA和2,4-D对怀牛膝愈伤组织生长的影响,并在愈伤组织形成高峰期30d时,采用酚-硫酸比色法对其中多糖的量进行检测,对所得实验数据进行极差分析、方差分析比较。结果大量元素的添加量对愈伤组织的诱导、生长和多糖的量无显著影响,6-BA对怀牛膝愈伤组织多糖量影响影响显著。结论诱导怀牛膝愈伤组织形成多糖的适宜培养基是1/4MS+6-BA0.5mg/L+2,4-D1.0mg/L。
Objective To study the effects of macronutrients and plant growth materials on the callus growth of Achyranthes bidentata and the amount of Polysaccharides in Achyranthes bidentata. Methods The leaves and stem segments of Achyranthes bidentata seedlings were used as material. The orthogonal experiment design method was used to study the effects of a large number of elements in the medium and the plant growth substances 6-BA, IBA, NAA and 2,4-D on Achyranthes bidentata. The influence of tissue growth and the 30-day peak period of callus formation were determined by phenol-sulfuric acid colorimetric method. The obtained experimental data were analyzed by range analysis and variance analysis. Results The amount of macroelement added had no significant effect on callus induction, growth and polysaccharide content. 6-BA had a significant effect on the polysaccharide content of callus from Achyranthes bidentata. Conclusion The suitable medium for inducing polysaccharide formation from callus of Achyranthes bidentata is 1/4MS+6-BA 0.5 mg/L+2,4-D 1.0 mg/L.
