目的高分离度快速液相色谱质谱联用(UFLC-QTRAP-MS/MS)同时测定不同组织部位和加工方法萝藦中15种氨基酸和12种核苷类成分的方法。方法采用Waters XBridge Amide色谱柱(2.1 mm×100 mm,3.5μm),以0.2%甲酸乙腈-0.2%甲酸水为流动相,梯度洗脱,流速0.6 m L·min~(-1),柱温为30℃,正离子多反应监测(MRM)模式测定。用灰色关联度分析(GRA)和TOPSIS分析进行综合性评价。结果 15种氨基酸成分和12种核苷成分在一定范围内线性关系良好,相关系数均大于0.997 3,平均加样回收率为98.17%~100.75%,相对标准偏差在0.90%~2.49%之间。不同部位和加工方法有一定的差异性,低温烘干的叶中氨基酸和核苷综合质量较好。结论该方法准确可靠,为选择最优处理方法提供可靠的方案。
OBJECTIVE To determine the contents of 15 kinds of amino acids and 12 kinds of nucleosides in different tissue parts and processing methods of Radish by high performance liquid chromatography-mass spectrometry (UFLC-QTRAP-MS / MS). Methods Waters XBridge Amide column (2.1 mm × 100 mm, 3.5 μm) was used with a mobile phase of 0.2% formic acid and 0.2% formic acid as mobile phase at a flow rate of 0.6 m L · min -1. The column temperature 30 ° C, positive ion multiple reaction monitoring (MRM) mode. The gray relational analysis (GRA) and TOPSIS analysis were used for comprehensive evaluation. Results The linear relationships of 15 kinds of amino acids and 12 kinds of nucleosides were good in a certain range. The correlation coefficient was higher than 0.997 3, the average recoveries were 98.17% -100.75%, and the relative standard deviations were between 0.90% and 2.49%. Different parts and processing methods have some differences, low-temperature dried leaves amino acids and nucleosides better overall quality. Conclusion The method is accurate and reliable, providing a reliable solution for selecting the optimal treatment method.
