目的 :观察 3种亲疏水性不同的新型药物辅料聚天冬酰胺衍生物PHPA ,PHPPA 3∶7和PHPPA 7∶3在动物体内的形态变化和降解过程。方法 :采用组织切片和高效液相凝胶色谱法 ,对小鼠的埋植部位、肝脏和肾脏进行观察和检测。结果 :发现3种材料在埋植部位均出现了从棕黑色固态到黄色胶状、再到棕色或黑色细小颗粒的形态变化 ,且其均能降解形成大小不同的分子片段。其中水溶性较强的PHPA较早出现细小颗料和降解高峰。三者均可被吸收到达肝肾组织。结论 :3种材料在小鼠皮下均可溶胀、降解 ,水溶性强的材料较早出现降解。
OBJECTIVE: To observe the morphological changes and degradation process of three kinds of new hydrophilic and hydrophilic hydrophobic drug derivatives, polyasparamanin derivatives, PHPA, PHPPA 3: 7 and PHPPA 7: 3. Methods: Tissue sections and high performance liquid chromatography were used to observe and detect the site of implantation, liver and kidney in mice. RESULTS: The morphological changes of the three materials from solid brown to yellow colloidal and then to brown or black fine particles were observed at the site of implantation, and all of them could degrade to form molecular fragments with different sizes. One of the more water-soluble PHPA earlier appeared small particles and degradation peak. All three can be absorbed to liver and kidney tissue. CONCLUSION: All three materials can be swollen, degraded and water-soluble in the skin of mice subsided earlier.
