目的:探讨MMP2基因在大肠癌肝脏转移中的意义。方法:收集手术切除21例大肠癌合并肝脏转移患者的原发病灶、癌旁正常黏膜组织、肝转移病灶及其周边正常肝组织的新鲜组织标本,用实时定量RT-PCR法测定各标本的MMP2 mRNA表达,并用Western blot方法检测其蛋白表达。结果:在肝转移灶、肝转移灶周边正常肝组织、大肠癌原发灶和正常大肠黏膜之间,全部病例的MMP2 mRNA表达均有明显差异,但以肝转移灶表达最高,正常大肠粘膜表达最低。其中18例在正常大肠黏膜中的MMP2蛋白表达低于肝转移灶、肝转移灶周边正常肝组织和原发灶。而所有病例原发灶与肝转移灶中的MMP2蛋白表达无差异,并且这二者的MMP2蛋白表达均高于肝转移灶周边正常肝组织。结论:MMP2在肝转移灶和原发灶中表达增强,有利于肿瘤细胞对基质的降解及其迁移。MMP2基因上调使得肝转移病灶中MMP2 mRNA的表达高于大肠癌原发病灶,可能是肝转移的分子机制之一。而肝转移灶周边正常肝组织中MMP2表达增强,可能与肝脏再转移有关。
Objective: To investigate the significance of MMP2 gene in liver metastasis of colorectal cancer. Methods: Fresh tissue samples from 21 primary colorectal cancer patients with liver metastasis, normal mucosa adjacent to liver cancer, liver metastasis and normal liver tissue from patients with colorectal cancer were collected. Real-time quantitative RT-PCR was used to detect MMP2 The mRNA expression was detected by Western blot. Results: The expressions of MMP2 mRNA in all the cases were significantly different between the liver metastases, the normal liver tissues around the liver metastases, the primary colorectal cancer and the normal colorectal mucosa. However, the expression of MMP2 mRNA in liver metastases was the highest but the expression in normal colorectal mucosa lowest. Among them, the expression of MMP2 protein in 18 cases of normal colorectal mucosa was lower than that in liver metastases, normal liver tissues and primary tumor around liver metastases. However, there was no difference in the expression of MMP2 protein between primary and hepatic metastases in all cases, and the expression of MMP2 protein in both of them was higher than that in the normal liver tissues around the liver metastases. Conclusion: The expression of MMP2 is enhanced in liver metastasis and primary tumor, which is beneficial to the degradation of tumor cells and their migration. Up-regulation of MMP2 gene makes the expression of MMP2 mRNA in liver metastasis higher than that of primary colorectal cancer, which may be one of the molecular mechanisms of liver metastasis. However, the expression of MMP2 in the normal liver tissue around the liver metastasis increased, which may be related to the liver metastasis.
