目的:观察糖肾汤对氧化低密度脂蛋白(ox-LDL)诱导的系膜细胞血凝素样氧化低密度脂蛋白受体1(LOX-1)表达及转化生长因子-β1(TGF-β1)和细胞外基质分泌的影响,探讨其防治糖尿病肾病的机制。方法:培养大鼠系膜细胞,分为空白组、ox-LDL组、糖肾汤高、中、低剂量组和罗格列酮组,培养24h后,RT-PCR检测系膜细胞mRNA表达,ELISA技术检测细胞上清液中TGF-β1、四型胶原(ColⅣ)和纤黏连蛋白(FN)含量。结果:糖肾汤、罗格列酮含药血清均能抑制ox-LDL诱导的TGF-β1、LOX-1 mRNA表达上调,及TGF-β1、FN、ColⅣ的分泌,其中糖肾汤含药血清呈剂量依赖性作用,中浓度作用最强。结论:糖肾汤可能通过降低ox-LDL受体LOX-1的表达以减少ox-LDL摄入及由此引起的细胞因子分泌和细胞外基质产生,从而起到防治糖尿病肾病的作用。
Objective: To observe the effect of Tangshen Decoction on the expression of LOX-1 and the level of TGF-β1 (TGF-β1) induced by ox-LDL in mesangial cells ) And extracellular matrix secretion to explore its mechanism of prevention and treatment of diabetic nephropathy. Methods: Rat mesangial cells were cultured and divided into blank group, ox-LDL group, high, medium and low dosage of Tangshen Tablet and rosiglitazone group. After cultured for 24h, mRNA expression of mesangial cells was detected by RT- ELISA was used to detect the content of TGF-β1, Col Ⅳ and FN in the supernatant. Results: Both Tangshen Decoction and Rosiglitazone-containing serum could inhibit the up-regulation of TGF-β1 and LOX-1 mRNA and the secretion of TGF-β1, FN and ColⅣ induced by ox-LDL, A dose-dependent effect, the strongest effect in the concentration. Conclusion: Tangshen Decoction may play a role in prevention and treatment of diabetic nephropathy by decreasing the expression of ox-LDL receptor LOX-1 to reduce the ox-LDL uptake and the production of cytokines and extracellular matrix.
