目的 应用前列腺梭形细胞瘤细胞系PC 981 0 6建立人前列腺肿瘤原位移植模型 ,观察肿瘤局部浸润和远处转移情况。方法 8只裸鼠取下腹部切口 ,显露膀胱和前列腺 ,将PC 981 0 6细胞悬液 0 .2~ 0 .4ml注射于前列腺后叶包膜下。观察裸鼠生命体征的变化 ,取原位肿瘤、膀胱、盆腔淋巴结、肺脏、肝脏、脾脏标本福马林液固定 48h ,苏木素 伊红染色后显微镜下观察。原位移植瘤电镜观察 ,X线照射显示骨骼转移情况。结果 5只模型建立成功 ,1 0d左右 ,可触及前列腺部位有小结节。 60d左右 ,裸鼠出现明显恶液质。解剖后发现盆腔淋巴结肿大 ,显微镜观察膀胱有浸润 ,肝脏有微转移灶。X线未见骨骼转移。结论 前列腺肿瘤原位移植模型的转归接近于临床 ,是肿瘤转移研究的较好模型
Objective To establish an orthotopic implantation model of human prostate tumor by using the cell line Prostate spindle cell line PC 981 0 6 to observe the local infiltration and distant metastasis of the tumor. Methods Eight nude mice were removed abdominal incision, the bladder and prostate were exposed, PC12000 cell suspension was injected into the posterior capsule of the posterior capsule. The changes of vital signs in nude mice were observed. The tumors in situ, bladder, pelvic lymph nodes, lung, liver and spleen specimens were fixed with formalin for 48 hours and stained with hematoxylin and eosin for microscopic observation. In situ xenograft electron microscopy, X-ray showed skeletal metastases. Results Five models were established successfully, around 10 days, there were small nodules in the reach of the prostate. About 60d, nude mice showed obvious cachexia. Anatomy found pelvic lymph node enlargement, microscopic observation of bladder infiltration, micrometastasis in the liver. No X-ray bone metastases. Conclusions The outcome of orthotopic transplantation of prostate tumor is closer to clinic and a better model of tumor metastasis
