目的:探讨吉西他滨常规化疗对人肝癌细胞株MHCC97H中肝癌干细胞样细胞的影响。方法:用免疫磁珠分选MH-CC97H中肝癌干细胞样细胞,将其接种到20只裸鼠皮下,建立移植瘤模型。将其随机分为治疗组和对照组(n=10)。治疗组:给予吉西他滨100mg/kg腹腔注射化疗(d1、d8、d15);对照组:在相同的时间内给予等量生理盐水。治疗后比较两组移植瘤大小变化;免疫组化检测肿瘤组织中CD133表达;流式细胞仪检测原代培养中CD133+细胞百分数。结果:治疗后,治疗组的移植瘤明显缩小,两组差异有统计学意义(P=0.046,P=0.012);治疗组和对照组免疫组化检测的灰度值分别为115.27±18.14和169.35±26.78,两组差异有统计学意义,P=0.041;治疗组和对照组原代培养中CD133+细胞百分数分别为(36.12±3.85)%和(14.65±1.98)%,两组差异有统计学意义,P=0.009。结论:肝癌干细胞样细胞具有多药耐药性,吉西他滨常规化疗对其没有杀伤作用。
Objective: To investigate the effect of gemcitabine routine chemotherapy on hepatoma stem cell-like cells in human hepatoma cell line MHCC97H. Methods: Liver cancer stem cell-like cells in MH-CC97H cells were sorted by immunomagnetic beads and inoculated into 20 nude mice subcutaneously to establish a transplanted tumor model. They were randomly divided into treatment group and control group (n = 10). Treatment group: gemcitabine 100mg / kg intraperitoneal injection chemotherapy (d1, d8, d15); control group: the same time given an equal amount of saline. After treatment, the size of the transplanted tumor was compared between the two groups; the expression of CD133 in the tumor tissue was detected by immunohistochemistry; the percentage of CD133 + cells in the primary culture was detected by flow cytometry. Results: After treatment, the transplanted tumor in the treatment group was significantly reduced, the difference between the two groups was statistically significant (P = 0.046, P = 0.012); the gray value of the immunohistochemistry in the treatment group and the control group were 115.27 ± 18.14 and 169.35 ± 26.78, the difference between the two groups was statistically significant (P = 0.041). The percentages of CD133 + cells in the primary culture and the control group were (36.12 ± 3.85)% and (14.65 ± 1.98)%, respectively, with significant difference between the two groups , P = 0.009. Conclusion: HCC stem cell-like cells have multidrug resistance, and gemcitabine has no cytotoxic effect by conventional chemotherapy.
