为快速、准确的对现有新疆野苹果种下类型和栽培品种进行鉴定,构建新疆苹果资源的DNA指纹图谱数据库。本研究利用SSR分子标记技术,先筛选鉴别能力强的SSR特征引物,再应用组合法构建62份新疆苹果资源DNA指纹图谱,最终对新疆野苹果种下类型和部分栽培品种进行聚类分析。研究显示,使用12对引物进行PCR扩增,共扩增条带80条,其中多态性条带79条,多态性百分率98.75%,筛选出鉴别能力最强的4对特征引物CH03d07、MS06g03、CH02a08、CH02b12;应用组合4对引物对62份新疆苹果种质资源构建指纹遗传图谱,能有效区分所有供试材料;同时根据该图谱的特异性标记位点,在遗传相似系数0.62处可将62个供试材料分为4大类群。从分析结果看,本研究构建的SSR标记适于构建新疆苹果资源的DNA指纹图谱库。
In order to quickly and accurately identify the existing cultivars and cultivars of wild apple in Xinjiang, DNA fingerprinting database of apple resources in Xinjiang was constructed. In this study, SSR markers were used to screen SSR primers with good discriminating ability. 62 DNA samples of Xinjiang apple were constructed by combinatorial analysis. Finally, cluster analysis was conducted on the types and partial cultivars of wild apple. The study showed that 12 pairs of primers were used for PCR amplification, and a total of 80 bands were amplified, of which 79 were polymorphic bands with a percentage of 98.75%. Four pairs of characteristic primers CH03d07 and MS06g03 , CH02a08 and CH02b12. The fingerprinting genetic map of 62 Xinjiang apple germplasm resources was constructed by primer pairs, which could effectively distinguish all the tested materials. According to the specific marker site of the map, genetic similarity coefficient of 0.62 62 test materials are divided into four groups. From the analysis results, the SSR markers constructed in this study are suitable for constructing a database of DNA fingerprinting of apple resources in Xinjiang.
