目的:观察甲状旁腺激素(human parathyroid hormone,PTH)对去卵巢大鼠牙槽骨的影响。方法:选用60只5月龄雌性大鼠随机分3组,其中两组作为药物治疗组和对照组均行全麻手术去除卵巢(ovariectomized,OVX),建立骨质疏松模型,另一组假手术组(sham-ovariectomized,sham-OVX)作为空白对照。6周后药物治疗组(PTH组)每日皮下注射人重组甲状旁腺激素(1-34)[recombinant human parathyroid hormone,rhPTH(1-34)],模型组(OVX组)和空白对照组(sham-OVX组)每日注射定量生理盐水,40 d后处死各组大鼠制取下颌骨标本。分析X线投照影像比较各组牙槽骨丧失程度;观察第一磨牙周围牙槽骨组织形态;检测牙槽骨破骨细胞数目进行比较。结果:影像学显示OVX组第一磨牙远中牙槽骨丧失(1.34±0.89)mm多于PTH组骨丧失量(0.78±0.62)mm和sham-OVX组骨丧失量(0.67±0.54)mm,差异具有统计学意义(P均<0.05);组织形态学染色显示OVX组牙槽骨骨质吸收较PTH组和sham-OVX组骨质吸收多;破骨细胞计数显示OVX组破骨细胞数(38.6±20.2)个分别多于PTH组(23.3±13.0)个和sham-OVX组(25.0±11.4)个,差异具有统计学意义(P均<0.01)。结论:PTH对骨质疏松大鼠牙槽骨的吸收有抑制作用。
Objective: To observe the effect of parathyroid hormone (PTH) on alveolar bone in ovariectomized rats. Methods: Sixty five-month-old female rats were randomly divided into three groups. The two groups were treated with general anesthesia for ovariectomized ovariectomized (OVX) to establish the model of osteoporosis. Another group of sham operation Sham-ovariectomized (sham-OVX) served as a blank control. After 6 weeks, the patients in the PTH group were injected subcutaneously with recombinant human parathyroid hormone (1-34) [rhPTH (1-34)], the model group (OVX group) and the blank control group sham-OVX group) were injected daily with normal saline. After 40 days, the rats in each group were sacrificed and the mandibular specimens were obtained. X-ray projection images were analyzed to compare the loss of alveolar bone in each group. The morphology of alveolar bone around the first molar was observed. The number of alveolar bone was detected for comparison. RESULTS: Imaging revealed more distal alveolar bone loss (1.34 ± 0.89) mm in OVX group than in PTH group (0.78 ± 0.62) mm and sham-OVX group (0.67 ± 0.54) mm, The difference was statistically significant (all P <0.05). The histological staining showed that the bone resorption of alveolar bone in OVX group was more than that in PTH group and sham-OVX group. The number of osteoclasts in OVX group 38.6 ± 20.2) were more than PTH group (23.3 ± 13.0) and sham-OVX group (25.0 ± 11.4) respectively, the difference was statistically significant (all P <0.01). Conclusion: PTH can inhibit the absorption of alveolar bone in rats with osteoporosis.
