[目的]观察沙立度胺(TD)及其对映体(S-TD)对人肺腺癌A549细胞增殖及对VEGF、MMP-9蛋白分泌的影响。[方法]体外培养A549细胞,采用MTT比色法,测定不同质量浓度的TD,S-TD对A549细胞增殖的影响;双抗体夹心酶联免疫吸附法(ELISA)测定细胞培养液中的VEGF和MMP-9蛋白含量。[结果]2.0~200.0μg/mlTD、S-TD作用48h,对A549细胞增殖无显著抑制作用(P>0.05);200.0μg/mlTD、S-TD对A549细胞VEGF蛋白分泌量有抑制活性作用,100.0~200.0μg/ml对MMP-9蛋白分泌具有抑制作用。[结论]TD及S-TD在体外对人肺腺癌A549细胞没有明显的增殖抑制作用;TD及S-TD在高浓度时抑制VEGF、MMP-9蛋白的分泌从而抑制肿瘤的增殖,但TD及S-TD活性无差异。
[Objective] To observe the effects of thalidomide (TD) and its enantiomer (S-TD) on the proliferation of human lung adenocarcinoma A549 cells and the secretion of VEGF and MMP-9. [Methods] The A549 cells were cultured in vitro. MTT colorimetric assay was used to determine the effect of different concentrations of TD and S-TD on the proliferation of A549 cells. The levels of VEGF and TGF-β1 were measured by double antibody sandwich enzyme-linked immunosorbent assay (ELISA) MMP-9 protein content. [Result] The proliferation of A549 cells was inhibited by 2.0 ~ 200.0μg / ml TD and S-TD for 48h, but no significant difference was found between the proliferation of A549 cells treated with 200 ~ 100.0 ~ 200.0μg / ml has an inhibitory effect on the secretion of MMP-9. [Conclusion] TD and S-TD have no obvious proliferation inhibitory effect on human lung adenocarcinoma A549 cells in vitro. TD and S-TD inhibit the secretion of VEGF and MMP-9 protein at high concentration and inhibit the proliferation of tumor cells. TD There was no difference in S-TD activity.
