目的:探讨重组人骨形成蛋白-2(rhBMP-2)和地塞米松(Dex)联合作用对牙囊细胞(DFCs)增殖和分化的影响。方法:利用四唑盐比色法(MTT),细胞总蛋白考马斯亮蓝测定法,碱性磷酸酶(ALP)试剂盒分别测定不同浓度的rhBMP和Dex单独和联合作用后DFCs的增殖和分化情况。结果:rhBMP对DFC的增殖和ALP的表达均有促进作用,低浓度Dex促进ALP的表达,但高浓度的Dex对DFC有增殖抑制作用,rhBMP和Dex联合作用后DFCs的增殖和ALP活性较单独作用有更明显的升高。结论:rhBMP和Dex联合作用对于促进DFCs的增殖和向成骨细胞分化有协同作用。
Objective: To investigate the effect of combined rhBMP-2 and Dex on the proliferation and differentiation of dental follicle cells (DFCs). Methods: The proliferation and differentiation of DFCs were measured by MTT assay, Coomassie brilliant blue assay and alkaline phosphatase (ALP) kit, respectively, with different concentration of rhBMP and Dex alone or in combination . Results: rhBMP could promote the proliferation of DFC and the expression of ALP, while Dex could promote the expression of ALP. However, high concentration of Dex could inhibit the proliferation of DFC, and the proliferation and ALP activity of DFC after rhBMP and Dex were more The role of a more significant increase. Conclusion: The combination of rhBMP and Dex has a synergistic effect on promoting the proliferation of DFCs and differentiating into osteoblasts.
