为研究从金丝小枣中提取类黄酮的最佳方法及该提取物的抗氧化活性,采用四因素三水平正交试验方案,研究水浴法和超声法提取类黄酮的最佳条件,并对这两种条件进行比较。通过测定类黄酮清除1,1-二苯代苦味酰基自由基(DPPH)能力和铁还原能力(FRAP),分析金丝小枣提取物中类黄酮的抗氧化活性。试验结果表明:水浴法:在料液比为1:50的条件下,用60%乙醇70℃提取2.5h,提取2次,类黄酮含量为10.25mg/g;超声法:在料液比为1:40的条件下,用60%乙醇,超声功率400W,提取40min,提取2次,类黄酮含量为11.08mg/g。抗氧化试验表明:金丝小枣类黄酮具有一定的抗氧化能力,其抗氧化活性随浓度的升高而增强。
In order to study the best method of extracting flavonoids from Ziziphus jujuba Mill. And the antioxidant activity of the extract, four-factor and three-level orthogonal experiment scheme was used to study the optimal conditions of extracting flavonoids by water bath and ultrasonic method. These two conditions are compared. Antioxidant activities of flavonoids in extracts of Ziziphus jujuba were determined by measuring the ability of flavonoids to scavenge 1, 1-dibenzyl picryl radical (DPPH) and iron reducing ability (FRAP). The results showed that: water bath method: the ratio of material to liquid was 1:50, extracted with 60% ethanol at 70 ℃ for 2.5 h, extracted twice, flavonoid content of 10.25 mg / g; ultrasonic method: 1:40 under the conditions of 60% ethanol, ultrasonic power 400W, extracted 40min, extracted twice, the flavonoid content of 11.08mg / g. Antioxidant tests showed that the flavonoids of Zizyphus jujuba cv. Ziziphus jujuba Mill. Had some antioxidant capacity and its antioxidant activity increased with the increase of its concentration.
