目的检测胆囊癌中凋亡抑制因子Survivin的表达,研究其与胆囊癌临床特征的关系,并探讨其与p53的相关性。方法采用免疫组织化学技术检测45例胆囊癌组织、20例胆囊息肉、20例胆囊炎及10例正常胆囊组织中Survivin和p53表达。结果45例胆囊癌组织中40例表达Survivin蛋白,34例表达p53蛋白,表达p53的病例中,31例表达Survivin阳性。而在胆囊息肉、胆囊炎及正常胆囊组织中不表达。且Survivin的表达与胆囊癌病人性别、年龄、肿瘤大小、病理分级、Nevin分期、淋巴结转移无关(P>0.05),而p53与淋巴结转移密切相关(P<0.01)。结论Survivin在胆囊癌中的高表达提示其可能与肿瘤的发生有关,Survivin和p53突变对凋亡抑制的协同作用在胆囊癌的发生发展过程中起重要作用。
Objective To detect the expression of Survivin, an inhibitor of apoptosis in gallbladder carcinoma (GCC), and to investigate its relationship with the clinical features of gallbladder carcinomas and its correlation with p53. Methods The expression of Survivin and p53 in 45 cases of gallbladder carcinoma, 20 cases of gallbladder polyps, 20 cases of cholecystitis and 10 cases of normal gallbladder tissues were detected by immunohistochemistry. Results Forty-five cases of gallbladder carcinoma expressed Survivin protein and 34 cases expressed p53 protein. Of the 45 cases with p53 expression, 31 cases expressed Survivin protein. In gallbladder polyps, cholecystitis and normal gallbladder tissue is not expressed. The expression of Survivin was not related to the gender, age, tumor size, pathological grade, Nevin stage and lymph node metastasis in patients with gallbladder carcinoma (P> 0.05), but p53 was closely associated with lymph node metastasis (P <0.01). Conclusions The high expression of Survivin in gallbladder carcinomas suggests that it may be related to tumorigenesis. The synergistic effect of Survivin and p53 mutations on apoptosis may play an important role in the carcinogenesis and progression of gallbladder carcinomas.
