目的:分析肿瘤细胞不同程度表达HLA-G对NK细胞杀伤活性影响。方法:以HepG2、NCI-H446、K562为研究对象,基因克隆建立表达载体,并将HLA-G/pGEM-T质粒转染到HepG2、NCI-H446及K562上。测定HepG2、NCI-H446及K562细胞内及细胞表面HLA-G的表达。结果:转染后的HEPG2-G、NcI-H446-G、K562-G细胞表面HEPG2-G的表达量依次为33.25%、45.68%、38.43%。3组比较,差异具有统计学意义(P<0.05)。
OBJECTIVE: To analyze the effect of HLA-G on NK cell cytotoxicity induced by tumor cells to varying degrees. Methods: HepG2, NCI-H446 and K562 were used as the research object and the gene clones were used to construct the expression vector. The plasmids were transfected into HepG2, NCI-H446 and K562 cells. The expression of HLA-G in HepG2, NCI-H446 and K562 cells and on the cell surface was determined. Results: The expression of HEPG2-G on HEPG2-G, NcI-H446-G and K562-G cells after transfection was 33.25%, 45.68% and 38.43%, respectively. Three groups, the difference was statistically significant (P <0.05).
