为了进一步探讨环胞霉素A (CSA)逆转白血病细胞耐药的机理 ,采用溴化四氮唑兰法(MTT)测定K5 62和K5 62 /ADM白血病细胞系对化疗的体外反应性 ,以脱氧核苷酸末端转移酶介导的缺口末端标记法 (TUNEL)及琼脂糖凝胶电泳检测细胞凋亡 ,台盼蓝拒染法测细胞存活率。结果发现CSA在 1.0mg/L水平对两种白血病细胞存活无明显影响 ,但能够增强其对化疗药物的敏感性 ,促进化疗药物诱导的白血病细胞凋亡 ,当浓度大于 1.0mg/L时单独CSA也能杀伤白血病细胞并诱导其凋亡。结论提示 ,CSA可以通过促进细胞凋亡逆转白血病细胞的耐药 ,其自身也有一定的诱导白血病细胞凋亡及杀伤白血病细胞的作用
To further investigate the mechanism of CSA reversing the drug resistance of leukemia cells, the in vitro reactivity of chemotherapy with K562 and K562 / ADM leukemia cell lines was determined by MTT assay. Apoptotic cells were detected by TUNEL and agarose gel electrophoresis. Cell viability was assayed by trypan blue exclusion. The results showed that CSA at 1.0mg / L had no significant effect on the survival of the two leukemic cells, but increased the sensitivity to chemotherapeutics and promoted chemotherapeutic drug-induced apoptosis of leukemia cells. When the concentration of CSA was above 1.0mg / L, CSA alone It can also kill leukemia cells and induce their apoptosis. Conclusions: CSA can reverse the drug resistance of leukemic cells by promoting apoptosis, and itself has a certain role in inducing leukemia cell apoptosis and killing leukemia cells
