目的建立吉祥草中凯提皂苷元的含测定方法。方法采用高效液相色谱(HPLC)-蒸发光散射检测器(ELSD),Agi-lent Ecplise XDB-C18(4.6mm×150mm,5μm)柱,甲醇-水(50∶50)为流动相,流速为1.0mL.min-1,柱温25℃,载气压力0.33MPa,漂移管温度65℃。结果凯提皂苷元质量浓度在0.1~1.5mg·mL-1之间线性关系良好,r=0.9995,平均回收率98.4%,RSD=1.5%(n=3)。结论该方法分离效果好,操作简便,稳定性好,重复性好,可用于吉祥草中凯提皂苷元的含测定,为吉祥草药材的开发利用奠定基础。
Objective To establish a method for the determination of triptolide in auspicious grass. Methods High performance liquid chromatography (HPLC) with evaporative light scattering detector (ELSD), Agi-lent Ecplise XDB-C18 column (4.6mm × 150mm, 5μm) and methanol-water (50:50) 1.0mL.min-1, column temperature 25 ℃, carrier gas pressure 0.33MPa, drift tube temperature 65 ℃. Results The linearity of the concentration of clenbuterol between 0.1 ~ 1.5mg · mL-1 was good, r = 0.9995, the average recovery was 98.4% and the RSD was 1.5% (n = 3). Conclusion The method has the advantages of good separation effect, simple operation, good stability and good repeatability. It can be used for the determination of trimethoprim-sulfamethoxazole in the auspicious grass, laying a foundation for the development and utilization of the auspicious herb.
