目的:探讨肝纤维化组织学量化诊断方法。方法:用二甲基亚硝胺(DMN)诱导SD大鼠肝纤维化模型,常规肝组织切片、Masson三色和苦味酸-天狼红染色,苦味酸固绿-天狼红染色比色测定,图像定量处理和半定量计分分析及羟脯氨酸(Hyp)含量测定,结果:切片胶原比色测定、图像定量处理和半定量计分分析均与肝纤维化程度变化相一致,与肝组织Hyp含量具有良好的相关性,r分别为0.94、0.89和0.88,P均<0.01。结论:切片胶原测定、图像定量处理和半定量计分可作为确定肝纤维化程度的量化指标。
Objective: To investigate the method of quantitative diagnosis of liver fibrosis. Methods: The model of hepatic fibrosis in SD rats was induced by dimethylnitrosamine (DMN). The liver tissues were stained with Masson’s trichrome and picric acid-Sirius red staining and the picric acid solid green-Sirius red staining , Quantification and semi-quantitative scoring analysis and hydroxyproline (Hyp) content determination. Results: The sections of collagen colorimetry, image quantification and semi-quantitative scoring analysis were consistent with the changes of liver fibrosis, Tissue Hyp content has a good correlation, r were 0.94,0.89 and 0.88, P <0.01. Conclusion: Slice collagen assay, image quantification and semi-quantitative scoring can be used as quantitative indicators to determine the degree of liver fibrosis.
