目的采用多指标正交实验考察黄芪有效部位黄芪黄酮、黄芪多糖、黄芪皂苷对糖尿病模型大鼠的药效机制,分析它们之间的交互关系。方法用正交设计安排实验,将126只大鼠随机分为9组,每组14只。除正常组外,其余各组按52 mg/kg剂量对大鼠进行一次性腹腔注射链脲佐菌素。造模后正常组、模型组给予蒸馏水6.3 mL/kg,其余各给药组分别按照0.105、0.284、0.389、0.043、0.148、0.327、0.432 g/kg剂量喂饲黄芪有效部位药物。造模后30 d测定血糖、血脂、脂联素、白细胞介素等药效、机制、指标。有关联的多项指标计算综合评分,对综合指标进行正交设计方差分析。结果模型组与正常组比较,除低密度脂蛋白外其他各项指标差异均有统计学意义(P<0.05)。黄酮有降血糖作用,多糖、皂苷作用不明显;黄酮、皂苷有降血脂作用,联合用药效果不如单一用药;黄酮、皂苷对脂联素有提升作用,二者合用效果优于单一用药。黄酮、多糖、皂苷对血清白介素有调节作用,黄酮和多糖、皂苷合用效果均优于单一用药。结论黄酮降血糖,黄酮、皂苷降血脂作用明显;在调节脂联素、白细胞介素方面,黄酮与皂苷具有协同作用。
Objective To investigate the pharmacodynamic mechanism of Astragalus membranaceus, Astragalus polysaccharide and Astragalus saponin in diabetic rats by multi-index orthogonal experiment and to analyze their interaction. Methods The experiments were designed by orthogonal design. 126 rats were randomly divided into 9 groups with 14 rats in each group. In addition to the normal group, the remaining groups were given a single intraperitoneal injection of streptozotocin at a dose of 52 mg / kg. After the model was established, the model group was given distilled water 6.3 mL / kg, and the rest of the drug-treated groups were fed with the active fractions of Astragalus according to the doses of 0.105,0.284,0.389,0.043,0.148,0.327,0.432 g / kg respectively. The blood glucose, blood lipid, adiponectin, interleukin and other pharmacodynamics, mechanism and index were measured 30 days after modeling. There are a number of indicators associated with the calculation of the comprehensive score, the comprehensive indicators of orthogonal design analysis of variance. Results Compared with the normal group, all the indexes except the low density lipoprotein in the model group had statistical significance (P <0.05). Flavonoids have hypoglycemic effects, polysaccharides, saponins effect is not obvious; flavonoids, saponins have hypolipidemic effect, combination therapy is not as good as a single drug; flavonoids, saponins on adiponectin have to enhance role, the combined effect is better than a single drug. Flavonoids, polysaccharides, saponins on the regulation of serum interleukin, flavonoids and polysaccharides, saponins were better than a single combination effect. Conclusions The anti-hyperlipidemia effect of flavonoids and flavonoids and saponins are obvious. In regulating adiponectin and interleukin, flavonoids and saponins have synergistic effect.
