利用基因枪法,以豇豆(Vigna aconitifolia)的Δ1-吡咯啉-5-羧酸合成酶基因(P5CS)的突变型P5CS-F129A基因转化高羊茅的下胚轴愈伤组织,获得25株经PCR检测和Southern b lot分析为阳性的转基因植株,转化频率约为1.4%。脯氨酸含量测定表明,转基因植株的脯氨酸含量比对照高31%~83%。对植株的抗旱性初步检测表明,转基因植株比对照耐旱。
The hypocotyl callus of Tall Fescue was transformed by the gene gun method using the mutant P5CS-F129A gene of Vigna aconitifolia’s Δ1-pyrroline-5-carboxylic acid synthase gene (P5CS) The transgenic plants tested positive for Southern b lot analysis had a transformation frequency of about 1.4%. Proline content determination showed that the proline content of transgenic plants was 31% ~ 83% higher than the control. Preliminary tests on the drought resistance of the plants showed that the transgenic plants were more drought tolerant than the control.
