采用AFLP技术对拟穴青蟹(Scylla paramamosain)子一代家系进行遗传学分析,并探讨了AFLP分子标记的遗传规律。32对ALFP选择性引物在拟穴青蟹家系G1代中共扩增出1 128个位点,其中不分离位点752个,多态性位点376个,多态位点比例为33.3%。在376个分离的位点中,符合孟德尔分离规律的位点共229个,占分离位点总数的60.9%;偏分离位点147个,占分离位点总数的39.1%。分离比为1∶1的位点共259个,占分离位点数的68.9%,其中符合孟德尔规律的位点155个,占59.8%;分离比为3∶1的位点共有117个,占分离位点数的31.1%,符合孟德尔分离规律的位点74个,占63.3%。32对引物组合检测到的有效等位基因个数在1.13-1.73之间;Shannon指数在0.10-0.56之间;遗传多样性指数在0.07-0.40之间。
AFLP technique was used to carry out genetic analysis of the pedigree of Scylla paramamosain and to investigate the genetic rules of AFLP markers. 32 pairs of ALFP selective primers amplified a total of 1 128 loci in the G1 generation of crab family, including 752 non-segregating loci, 376 polymorphic loci and 33.3% polymorphic loci. Among the 376 isolated sites, 229 sites met the Mendelian separation rule, accounting for 60.9% of the total number of isolated sites and 147 partial sites, accounting for 39.1% of the total number of isolated sites. A total of 259 isolates with a ratio of 1: 1 accounted for 68.9% of the total number of isolates, of which 155 were Mendelian-based, accounting for 59.8%. A total of 117 loci with a segregation ratio of 3: 1 accounted for Seventy-one percent of the sites were segregated, with 74 sites meeting the Mendel’s separation rule, accounting for 63.3%. The number of effective alleles detected by 32 pairs of primers was between 1.13-1.73, Shannon index was 0.10-0.56, and the genetic diversity index was between 0.07-0.40.
