目的:筛选出青黛炮制过程中显著影响靛玉红生成的因素,优化最佳水平组合,确定生成靛玉红的最佳工艺。方法:以鲜叶产靛玉红的质量分数(mg·g-1)为指标,采用Plackett-Burman设计,Box-Behnken设计效应面分析等统计学方法,筛选出显著影响因素及最佳水平组合。结果:青黛的浸泡、打靛工艺确定为浸泡前不除蜡、浸泡液pH7、溶剂用量-鲜叶质量(mL∶g)15、浸泡不避光、浸泡48h、温度60℃、通气时间180min、加氨水调节打靛前pH至10.5。结论:系统考察了青黛炮制过程中各个影响因素对产生的有效成分靛玉红的影响,使工业化生产控制靛玉红的生成变为现实,为进一步研究青黛炮制原理奠定了基础。
OBJECTIVE: To screen out the factors that significantly affect the production of indirubin during the processing of indigo, optimize the optimal combination of levels, and determine the best process for producing indirubin. Methods: According to the statistic method of Plackett-Burman design and Box-Behnken design effect surface analysis, the quality factors (mg · g-1) of fresh-producing indirubin were used as indexes to screen out the significant influencing factors and the optimal level combination . Results: The indigo process of indigo was determined as not wax removal before immersion, soaking liquor pH7, the amount of solvent-fresh leaf mass (mL: g) 15, soaking for 40h, soaking for 48h, temperature 60 ℃, aeration time 180min, Add ammonia to adjust the pH before playing to 10.5. Conclusion: The influence of various influencing factors on the indirubin, which is the active ingredient, was systematically investigated in the process of processing indigo, which made it possible to control the production of indirubin in industrialized production. This laid the foundation for further study on the processing principle of indigo.
