目的观察依那普利和中药复方对糖尿病大鼠肾小球丝裂原活化蛋白激酶 (MAPK) ,蛋白激酶Cα(PKCα) ,转化生长因子β1(TGFβ1)表达和纤维连接蛋白 (FN)沉积的影响。方法Wistar大鼠随机分为正常对照组 (A组 )和糖尿病组 (B、C、D组 )。用链脲菌素复制糖尿病模型 ,B组为糖尿病对照组 ,C组为糖尿病依那普利组 ,按 2mg/kg .d给药 ,D组为糖尿病中药组 ,按 1 8g/kg .d中药浸膏给药 ,持续 6 0天。用免疫组化检测MAPK、PKCα、TGFβ1和FN在肾小球的表达 ;用生化方法测定大鼠血糖、血脂、血肌酐和尿蛋白 ;光镜观察肾小球形态。结果糖尿病各组肾小球MAPK、PKCα、TGFβ1和FN的表达明显高于正常对照组 ,但用药组 (C、D组 )的表达则低于未用药组 (B组 )。形态观察显示 ,C、D组肾小球病变比B组轻。B组血糖与C、D组无明显差异 ,但血脂、血肌酐和尿蛋白则高于C、D组。结论依那普利和丹芪复方可抑制糖尿病大鼠肾小球MAPK和PKCα 信号途径 ,从而改善肾小球的形态和功能。
Objective To investigate the effects of enalapril and traditional Chinese medicine on the deposition of mitogen-activated protein kinase (MAPK), PKCα, TGFβ1 and FN in diabetic rats . Methods Wistar rats were randomly divided into normal control group (group A) and diabetic group (group B, C and D). Diabetic model was replicated with streptozotocin, group B was diabetic control group, group C was diabetic enalapril group, dosed as 2mg / kg.d group, group D was diabetes mellitus group, treated with 18g / kg.d medicine The extract is administered for 60 days. Immunohistochemistry was used to detect the expression of MAPK, PKCα, TGFβ1 and FN in glomeruli. The blood glucose, blood lipid, serum creatinine and urinary protein were determined by biochemical methods. The glomerular morphology was observed with light microscope. Results The expressions of MAPK, PKCα, TGFβ1 and FN in diabetic glomerulus group were significantly higher than those in normal control group (P <0.05). Morphological observation showed that glomerular lesions in C and D groups were lighter than those in B group. There was no significant difference between group B and group C and group D, but the levels of serum lipids, serum creatinine and urine protein were higher than those of group C and D. Conclusion Enalapril and Danqi compound can inhibit the signal pathways of MAPK and PKCα in glomeruli of diabetic rats and thus improve glomerular morphology and function.
