目的:研究过表达ALEX1对乳腺癌MCF-7细胞增殖和凋亡的影响。方法:将重组慢病毒LV5-ALEX1及阴性对照LV5-NC分别感染乳腺癌MCF-7细胞。感染72 h后,采用实时定量PCR和Western blot分别检测感染后ALEX1的表达情况;感染24、48、72、96 h,CCK8法检测细胞增殖情况;感染72 h后,流式细胞术检测细胞凋亡情况。结果:感染72 h后细胞ALEX1 mRNA表达水平明显上升(165.81±12.14 vs 52.29±2.32,P<0.01),实验组与对照组相比,细胞增殖受到明显抑制,细胞凋亡率明显增加(20.55%±2.50%vs 3.60%±1.614%,P<0.05)。结论:过表达ALEX1能明显抑制乳腺癌MCF-7细胞的增殖并诱导细胞发生凋亡。
Objective: To investigate the effect of overexpression of ALEX1 on proliferation and apoptosis of breast cancer MCF-7 cells. Methods: Breast cancer MCF-7 cells were infected with recombinant lentivirus LV5-ALEX1 and negative control LV5-NC respectively. After infection for 72 h, the expression of ALEX1 was detected by real-time PCR and Western blot, respectively. After 24, 48, 72 and 96 h of infection, the cell proliferation was detected by CCK8 assay. The apoptosis was detected by flow cytometry 72 h after infection Death situation. Results: The expression of ALEX1 mRNA was significantly increased at 72 h after infection (165.81 ± 12.14 vs 52.29 ± 2.32, P <0.01). Compared with the control group, the cell proliferation was significantly inhibited and the apoptosis rate was significantly increased (20.55% ± 2.50% vs 3.60% ± 1.614%, P <0.05). Conclusion: Overexpression of ALEX1 can significantly inhibit the proliferation of MCF-7 breast cancer cells and induce cell apoptosis.
