目的:为了了解血栓形成后血管内皮细胞释放内皮依赖性舒张因子(EDRF)的功能是否改变,实验观察了血栓形成对乙酰胆碱(Ach)内皮依赖性舒张血管作用的影响.方法:大鼠足底皮下局部注射0.5%角叉菜胶(carrageenan),制备大鼠尾动脉血栓形成模型.将血栓形成部位的血管制成离体血管条,置于充满37℃Krebs液的浴槽中,以1O ̄(-7)mol/L去甲肾上腺素(NE)引起血管条收缩,观察Ach和硝酸甘油的作用。结果:血栓形成开始后6~8h,Ach(10 ̄(-8),10 ̄(-7),10 ̄(-6)mol/L)剂量依赖性地舒张尾动脉血管条(24.8±3.3%,42.1±4.7%,66.6±3.8%,P<0.05);血栓形成开始后8~16h,Ach剂量依赖性地舒张血管条作用明显减弱(9.3±1.5%,15.9±2.9%,19.7±3.4%,P<0.001);血栓形成开始16h后,血管条对10 ̄(-7)mol/LNE的反应性消失;机械法擦去尾动脉内皮后,Ach剂量依赖性地舒张血管条的作用亦明显减弱(P<0.001),与血栓形成后8~16h时的Ach作用之间无显著差别(P>0.05).去内皮或血栓形成后(6~8h,8~?
