作者用体外短期药敏试验测定50例急性白血病对药物的敏感性。病人的白血病细胞分别与HH、Adr、Ara-C、VCR和Pred等培养2小时,接着加入~3H-UR继续培养1小时。药物的抑制作用以~3H-UR的掺入抑制百分率SI(%)来表示。对38例在体外和体内均用过HH的病人作了回顾性——相关性研究,发现如HH的SI>15%时,在体内敏感,<15%则抗药,体外与体内的相符率为87%(33/38).我们认为使用~3H-UR掺八的体外短期药敏试验比使用~3H-TdR更为可靠.在急性白血病化疗前可采用此法预测疗效.
The authors used in vitro short-term susceptibility testing of 50 cases of acute leukemia drug sensitivity. The leukemia cells of the patients were cultured with HH, Adr, Ara-C, VCR and Pred respectively for 2 hours, followed by addition of ~ 3H-UR for 1 hour. The inhibitory effect of the drug is expressed as the% inhibition of incorporation of 3H-UR (%). A retrospective-correlation study was performed in 38 patients with HH both in vitro and in vivo and found to be sensitive in vivo when SI> 15% of HH, <15% resistant, in vitro and in vivo compatible Was 87% (33/38) .We believe that the use of ~ 3H-UR mixed eight in vitro short-term drug susceptibility testing than the use of ~ 3H-TdR more reliable in the treatment of acute leukemia can be used to predict the efficacy of chemotherapy.
