目的:研究非小细胞肺癌(NSCLC)中磷酸化细胞外信号调节激酶(PERK1/2)及靶基因产物CyclinD1和血管内皮生长因子(VEGF)的表达关系,探讨PERK1/2在NSCLC发生、发展中的作用。方法:应用免疫组织化学技术检测10例正常肺组织和64例NSCLC组织中PERK1/2、CyclinD1和VEGF的表达情况,并与临床病理特征进行统计学分析。结果:NSCLC组织中PERK1/2、CyclinD1和VEGF的阳性表达率分别为57.8%、60.9%和71.9%,正常肺组织均为阴性表达;PERK1/2的核内表达和VEGF的表达率与TNM分期、淋巴结转移相关(P<0.05),CyclinD1的阳性表达率与肿瘤的分化程度及淋巴结转移相关(P<0.05);PERK1/2的核内表达与CyclinD1在NSCLC中的表达呈线性相关(P<0.05)。结论:PERK1/2介导的信号转导事件可能在NSCLC发生、发展过程中起重要促进作用。
Objective: To investigate the expression of phosphorylated extracellular signal-regulated kinase (PERK1 / 2) and its target gene products CyclinD1 and VEGF in non-small cell lung cancer (NSCLC) and to investigate the role of PERK1 / 2 in the occurrence and development of NSCLC Role. Methods: The expressions of PERK1 / 2, CyclinD1 and VEGF in 10 normal lung tissues and 64 NSCLC tissues were detected by immunohistochemistry and analyzed with clinicopathological features. Results: The positive rates of PERK1 / 2, CyclinD1 and VEGF in NSCLC tissues were 57.8%, 60.9% and 71.9%, respectively. The expression of PERK1 / 2 in nucleus and the expression of VEGF were correlated with TNM stage , And lymph node metastasis (P <0.05). The positive rate of CyclinD1 was correlated with tumor differentiation and lymph node metastasis (P <0.05). The expression of PERK1 / 2 in nucleus and the expression of CyclinD1 in NSCLC were linearly correlated (P < 0.05). CONCLUSION: PERK1 / 2-mediated signal transduction events may play an important role in the development and progression of NSCLC.
