经对浙江省甘蔗花叶病毒(ScMV)的甜玉米分离物的研究表明,其钝化温度(TIP)为55℃,稀释终点(DEP)为10~(-4)—10~(-5),体外保毒期(LIV)为48小时(20—25℃)。在部分高粱品种如Rio、Atlas、Bugoff,还有软三A品系上产生局部坏死斑,在玉米和苏丹草上为系统感染,但不侵染约翰逊草和大麦。初步认为该分离物中至少含有2个株系。 用TAC(0.1M Tris—0.05M柠檬酸)缓冲系统(pH 8.3)提取该分离物,再经氯仿澄清,然后配合低速离心(4000rpm)进行两次8%聚乙二醇(PEG6000或12000)浓缩。这样,部分纯化的样品在260nm O.D.值最大,230—240nm O.D.最小,其260/280:126±0.08;280/260:0.83±0.06。获得此抗原的血清效价:琼脂双扩散1/256,试管沉淀1/512,微量沉淀1/512。 毒源测试结果:与带毒种蔗,田间甘蔗、玉米、苏丹草病株组织有阳性反应,均可形成明显的沉淀带,而对照均为阴性反应。
The results showed that the TIP of ScMV was 55 ℃ and the DEP was 10 -4 -4 -10 -5. , In vitro protection period (LIV) for 48 hours (20-25 ℃). Local necrotic spots were produced on some sorghum varieties such as Rio, Atlas, Bugoff, and also on Soft Triple A lines, and were systematically infected on maize and sudangrass but not on Johnson grass and barley. It is preliminarily thought that this isolate contains at least 2 lines. The isolate was extracted with a TAC (0.1 M Tris-0.05 M citric acid) buffer system (pH 8.3) and then clarified by chloroform and then concentrated with 8% polyethylene glycol (PEG 6000 or 12000) twice with low speed centrifugation (4000 rpm) . Thus, the partially purified sample had the highest O.D. values at 260 nm, 230-240 nm O.D. min., 260/280: 126 + 0.08; 280/260: 0.83 + 0.06. The serum titer of this antigen: agar double diffusion 1/256, tube precipitation 1/512, trace precipitation 1/512. Toxicity Test Results: Positive reaction with poisonous sugarcane, field sugarcane, corn, Sudan grass plant tissue, the formation of a clear sedimentation zone, while the control were negative.
