目的研究磷脂酶A2(PLA2)抑制剂地塞米松(DXM)对内毒素血症动物的保护效应。方法动物分为两组,即内毒素血症组和地塞米松组。采用家兔内毒素血症模型,测定内毒素血症组、PLA2抑制剂DXM预处理组动物平均动脉压(MAP)、8小时活存率、血浆PLA2活性和丙二醛(MDA)含量。结果注射内毒素后MAP显著下降,8小时存活率为57%;血浆PLA2活性及MDA含量显著升高,并随着伤情的加重而升高。而DXM预处理组,注射内毒素后MAP下降幅度显著小于内毒素组(P<0.05),血浆PLA2活性及MDA含量也显著低于内毒素组,动物8小时活存率达70%。结论 PLA2抑制剂DXM能显著提高动物活存率和活存时间,改善血液动力学,抑制PLA2活性。提示DXM在抗休克和全身炎症方面有较好效果。“,”Objective To explore the protective effects of dexamethasone(DXM)-the inhibitor of phospholipase A2(PLA2) on endotoxemia animals.Methods The animals were divided into 2 groups, i.e, endotoxemia group and DXM group. A rabbit endotoxemia model was used to measure the mean arterial pressure (MAP), survival rate after 8 hours, PLA2 activity of plasma and MDA level of the endotoxemia group and DXM group.Results After injected with endotoxin, MAP was decreased significantly with a survival rate of 57% after 8 hours; however, PLA2 activity of plasma and MDA level were significantly increased along with the aggravation of injury. As for the DXM pretreatment group, after injected with endotoxin, MAP was decreased less significantly than that of the endotoxin group(P<0.05); the PLA2 activity of plasma and the MDA level were significantly lower than those of the endotoxin group. The survival rate after 8 hours was as high as 70%.Conclusion The inhibitor (DXM) of PLA2 can elevate animal survival rate and survival time, improve hemodynamics and depress PLA2 activity. It suggests that DXM has positive effect on shock and general inflammation.
