目的:观察补肾益智方对D-半乳糖加速衰老加上脑Meynert核损毁Alzheimer病(AD)模型大鼠脑内生长抑素(somatostatin, SS)的影响。方法:将15月龄初老年雌性Wistar大鼠随机分为正常对照组、AD模型组、双益平对照组和补肾益智方观察组。模型组大鼠以D-半乳糖腹腔注射加上鹅膏蕈氨酸(i-botenic acid,IBO)脑内Meynert核注射造模,双益平对照组和补肾益智方观察组分别在造模的同时灌胃双益平和补肾益智方。运用免疫组织化学和cRNA探针原位杂交方法检测大鼠大脑顶叶皮质、海马 CA_1和齿状回门区SS免疫和SSmRNA表达阳性神经元数目。结果:补肾益智方组大鼠脑顶叶、海马CA_1、齿状回门区SS免疫和SSmRNA表达阳性神经元的数目及其积分光密度较模型组有增加趋势,部分组间比较有显著性差异。结论:补肾益智方对D-半乳糖加上IBO损毁Meynert核AD模型大鼠脑内SS能神经元有一定程度的保护作用。
OBJECTIVE: To observe the effect of Bushen Yizhi Recipe on somatostatin (SS) in brain of Alzheimer disease (AD) rats induced by accelerated aging of D-galactose and brain Meynert nuclear damage. Methods: Female Wistar rats aged 15 months old were randomly divided into normal control group, AD model group, Shuangyiping control group and Bushen Yizhi group. The rats in the model group were injected intraperitoneally with D-galactose plus intrathecal Meynert nuclear injection of i-botenic acid (IBO). The Shuangyiping control group and the Bushen Yizhi group were observed in the model group. At the same time gavage Shuangyi Pinghe Bushen Yizhi Fang. Immunohistochemistry and cRNA probe in situ hybridization were used to detect the number of SS immunoreactive and SS mRNA-expressing neurons in the rat parietal cortex, hippocampus CA-1 and dentate gyrus. RESULTS: The number and integrated optical density of SS immunoreactive and SS mRNA-expressing neurons in the parietal lobe, hippocampal CA1, and dentate gyrus of rats in the Bushen Yizhi Recipe group were increased compared with the model group, and there was significant difference among the groups. difference. Conclusion: Bushen Yizhi Decoction has a certain degree of protective effect on the SS neurons in the brain of Meynert rats with D-galactose plus IBO.
