目的观察瘦素(Leptin)对肝细胞葡萄糖氧化的影响及其剂量效应,并观察其对葡萄糖激酶基因表达的影响。方法正常大鼠肝细胞,以不加Leptin与加不同剂量的Leptin培养1小时后,以液体闪烁计数法检测肝细胞氧化〔U14C〕葡萄糖的量。以RTPCR法检测Leptin100μg/L处理组与对照组肝细胞葡萄糖激酶(GK)mRNA的表达。结果当Leptin浓度为10μg/L时,肝细胞葡萄糖氧化与对照组无明显差异,当Leptin浓度上升为50μg/L后,葡萄糖氧化较对照组显著下降(P<0.05),并随剂量增大抑制作用加强。Leptin100μg/L处理组GKmRNA表达水平(以βactin校正)亦显著低于对照组(0.43±0.07比0.87±0.09,P<0.01)。结论低剂量瘦素对肝细胞葡萄糖氧化无明显影响,高浓度瘦素则具剂量依赖性抑制作用,并至少部分通过抑制GK基因表达而达到
Objective To observe the effects of leptin on glucose oxidation in hepatocytes and its dose effects and to observe its effect on glucokinase gene expression. Methods Normal rat hepatocytes were cultured for 1 hour with or without Leptin supplemented with different doses of Leptin. The amount of [U-14C] glucose oxidized by hepatocytes was measured by liquid scintillation counting. The expression of glucose kinase (GK) mRNA in hepatocytes of 100μg / L Leptin treated group and control group was detected by RTPCR. Results When the concentration of Leptin was 10μg / L, there was no significant difference between the two groups (P <0.05). When the concentration of Leptin increased to 50μg / L, the glucose oxidation decreased significantly compared with the control group (P <0.05) Large inhibition enhanced. The level of GK mRNA (corrected by β-actin) in Leptin 100μg / L group was also significantly lower than that in control group (0.43 ± 0.07 vs 0.87 ± 0.09, P <0.01). Conclusions Low-dose leptin has no effect on the glucose oxidation of hepatocytes. High-concentration leptin has a dose-dependent inhibitory effect and is at least partially achieved by inhibiting GK gene expression
