目的观察17β-雌二醇对大鼠心缺血再灌(I/R)损伤的保护作用并探讨其机制。方法30只成年雌性SD大鼠卵巢切除后,随机分成C组(I/R假手术组)、I/R组和I/R+E2组(17β-雌二醇预处理)。17β-雌二醇预处理2周后建立大鼠心肌缺血再灌注模型,用氯化三苯甲基四氮唑(TTC)染色法测量心肌梗死范围,流式细胞术检测心肌细胞凋亡率,免疫组织化学方法检测心肌细胞原癌基因cf-os的蛋白表达。结果17β-雌二醇能明显减少缺血再灌注心肌梗死体积,降低心肌细胞凋亡率,减弱缺血再灌注心肌细胞c-Fos的表达。结论雌激素对大鼠缺血再灌注心有保护作用,其作用机制可能与抑制原癌基因cf-os的蛋白表达有关。
Objective To observe the protective effect of 17β-estradiol on myocardial ischemia-reperfusion (I / R) injury in rats and its mechanism. Methods Thirty adult female Sprague-Dawley rats were randomly divided into C group (I / R sham operation group), I / R group and I / R + E2 group (17β-estradiol pretreatment). Myocardial ischemia-reperfusion model was established by 17β-estradiol preconditioning for 2 weeks. The extent of myocardial infarction was measured by TTC staining. The apoptosis rate of myocardial cells was detected by flow cytometry The protein expression of cf-os in cardiomyocytes was detected by immunohistochemistry. Results 17β-Estradiol significantly reduced the volume of myocardial infarction after ischemia-reperfusion, decreased the rate of cardiomyocyte apoptosis and decreased the expression of c-Fos in ischemia-reperfusion myocardium. Conclusion Estrogen has a protective effect on myocardial ischemia-reperfusion in rats, and its mechanism may be related to the inhibition of cf-os protein expression.
